Vascular Targeting Agent Halts Bone Metastasis
A novel vascular targeting agent completely prevented the development of bone tumors in 50 percent of the mice tested in a preclinical study, providing early evidence that it could treat, or thwart, growth of tumors in bone, a common destination for many cancers when they start to spread.

Scientists at The University of Texas M. D. Anderson Cancer Center published in the journal Cancer Research that this "Trojan Horse" agent, VEGF121/rGel, stopped specialized cells within the bone from chewing up other bone material to make room for the implanted tumor to grow.

Eventhough this study tested the ability of VEGF121/rGel to halt the growth of human prostate cancer cells in the bones of mice, researchers say it likely could help prevent the growth of other cancers in bones such as breast, multiple myeloma, lung and renal cell.

"A number of tumors invade bone in the same way, so these findings suggest it may be possible to shut down this process regardless of the tumor type," says the study's lead author, Michael G. Rosenblum, Ph.D., professor in the Department of Experimental Therapeutics. "If that could be done - and we are a long way from determining if it is possible - we may be able to offer the first therapy that specifically targets bone metastasis".........

Genes offer researchers a 'crystal ball'
The science of cancer prevention has advanced to the point where researchers now say they can detect "cancer genes" in the breath of smokers, and can test the presence of two proteins in men they say will predict development of prostate cancer a decade in advance. All of these novel findings need much more examination, of course, but scientists at the American Association for Cancer Research's Frontiers in Cancer Prevention Research meeting, say these examples illustrate how it is becoming increasingly possible to use genes and their protein products to help predict and diagnose cancer, as well as choose therapy that offers the most potential for a good result. These researchers will also discuss a test that can pick out patients who have pancreatic cancer - an advance that offers hope the disease can be treated at earlier stages than it is now - and how several unique genes can predict which prostate cancer or lung cancer patients will develop aggressive tumors that need additional treatment. Cancer is a disease of genes, they say, so genes can be employed as a crystal ball to thwart the disease.

Lung carcinogenesis tracked by DNA methylation mapping in exhaled breath.

For the first time, researchers have demonstrated that it is possible to detect DNA methylation in the breath of smokers and lung cancer patients, suggesting that, in theory, it may be possible to use this technique to identify people who have undiagnosed lung cancer or are at high risk of developing the disease.........

Vaccine Shows Promise Against Breast Cancer
A diagnosis of breast cancer has taken on a new meaning in the past 10 years, as research has produced a host of new therapies and detection techniques, significantly improving long-term survival for women who have been fighting the disease. To build on these successes, scientists are now harnessing what they have learned about treating breast cancer and applying it to possible methods of prevention to reduce the total occurence rate of the disease. One study presented today at the American Association for Cancer Research's Frontiers in Cancer Prevention Research meeting in Boston looks at a specific target in the fight against breast cancer and evaluates a potential vaccine that is yielding promising results for women who are at high-risk for the disease.

Targeted immunoediting of critical pathways responsible for breast cancer development: therapy of early breast cancer using HER-2/neu pulsed dendritic cells.

Multiple genetic targets have been discovered that may help fight breast cancer, including BRCA, estrogen receptors, and HER-2/neu, all of which have been known to predict the severity of disease, recurrence and overall survival. Developing novel therapies that target these specific genetic variances may be extremely beneficial in preventing breast cancer for a number of women.........

Peer And Family Support For Cancer Survivors
Adolescent and young adult cancer patients rank support from family, friends and other cancer survivors as high priority healthcare needs, according to a new University of Southern California study. Published in the recent issue of CANCER, a peer-reviewed journal of the American Cancer Society, the study reveals this traditionally underserved population of 15-29 year-old cancer survivors feels that socially connecting with other cancer-afflicted peers of the same age may in some cases be more beneficial than receiving support from family and friends, contrary to what their physicians believe.

Led by Brad Zebrack, Ph.D., M.S.W. of the University of Southern California School of Social Work in Los Angeles, researchers conducted a comprehensive survey with oncologists, psychologists, nurses, social workers and young adult cancer survivors to better characterize the needs of this patient population and rank them in terms of importance.

According to Dr. Zebrack, "health professionals and survivors value highly the support of family and friends. However, meeting other young people who share a common experience becomes an opportunity for young adult cancer patients and survivors to address common concerns, such as coping with uncertainty about one's health and future, feelings of being alone and isolated, body changes, sexuality and intimacy, dating and relationships, and employment issues".........

Firefighters Face Increased Risk Of Cancers
University of Cincinnati (UC) environmental health researchers have determined that firefighters are significantly more likely to develop four different types of cancer than workers in other fields.

Their findings suggest that the protective equipment firefighters have used in the past didn't do a good job in protecting them against cancer-causing agents they encounter in their profession, the researchers say.

The researchers found, for example, that firefighters are twice as likely to develop testicular cancer and have significantly higher rates of non-Hodgkin's lymphoma and prostate cancer than non-firefighters. The researchers also confirmed previous findings that firefighters are at greater risk for multiple myeloma.

Grace LeMasters, PhD, Ash Genaidy, PhD, and James Lockey, MD, report these findings in the November edition of the Journal of Occupational and Environmental Medicine. The UC-led study is the largest comprehensive study to date investigating cancer risk associated with working as a firefighter.

"We believe there's a direct correlation between the chemical exposures firefighters experience on the job and their increased risk for cancer," says LeMasters, professor of epidemiology and biostatistics at UC.

Firefighters are exposed to many compounds designated as carcinogens by the International Agency for Research on Cancer (IARC)--including benzene, diesel engine exhaust, chloroform, soot, styrene and formaldehyde, LeMasters explains. These substances can be inhaled or absorbed through the skin and occur both at the scene of a fire and in the firehouse, where idling diesel fire trucks produce diesel exhaust.........

With the help of tiny, transparent zebrafish embryos, scientists at the Kimmel Cancer Center at Thomas Jefferson University and Jefferson Medical College are hoping to prove that a microscopic nanoparticle can be part of a "new class of radioprotective agents" that help protect normal tissue from radiation damage just as well as standard drugs.

Reporting November 7, 2006 at the annual meeting of the American Society for Therapeutic Radiology and Oncology in Philadelphia, they show that the nanoparticle, DF-1 - a soccer ball-shaped, hollow, carbon-based structure known as a fullerene - is as good as two other antioxidant drugs and the FDA-approved drug, Amifostine in fending off radiation damage from normal tissue.

The scientists, led by Adam Dicker, M.D., Ph.D., professor of radiation oncology at Jefferson Medical College of Thomas Jefferson University in Philadelphia and at Jefferson's Kimmel Cancer Center, and Ulrich Rodeck, M.D., professor of dermatology at Jefferson Medical College, compared DF-1 to two superoxidase dismutase mimetics, which are antioxidant drugs. They exposed zebrafish embryos to radiation with either DF-1 or a sod or amifostine. Each of the three markedly reduced radiation damage and increased overall survival and was comparable to the protection provided by the Amifostine.........

Muscle Protein Drives Prostate Cancer
Scientists at the Johns Hopkins Kimmel Cancer Center have for the first time implicated the muscle protein myosin VI in the development of prostate cancer and its spread.

In a series of lab studies with human prostate cancer cells, the Hopkins researchers were surprised to find overproduction of myosin VI in both prostate tumor cells and premalignant lesions. When the researchers genetically altered the cells to "silence" myosin VI, they discovered the cells were less able to invade in a test tube.

"Our results suggest that myosin VI may be critical in starting and maintaining the cancerous properties of the majority of human prostate cancers diagnosed today," says Angelo M. De Marzo, M.D., Ph.D., a study coauthor and associate professor of pathology, urology and oncology.

The Hopkins work, reported in the recent issue of the American Journal of Pathology, has potential value for better ways to diagnose the disease, treat and track the effects of drugs and surgery. "Targeting myosin VI represents a promising new approach that could lead eventually new approaches to treating the disease," says Jun Luo, Ph.D., senior author of the paper and assistant professor of urology.

Myosins are a class of 40 motor proteins that power cell movement and muscle contractions. Normally, as they work, myosins slide in a single direction along the threads of a protein called actin. But myosin VI moves against the grain, and it does not function as a classical "muscle" protein.........

Smoking, Fractures And Ligament Injuries
Cigarette smoking, which causes over 8.6 million illnesses annually in the U.S., has been shown to have harmful effects on a variety of orthopedic conditions. Studies have shown that the numerous toxins contained in cigarette smoke can undermine fracture and ligament repair following injury. In addition, smokers have higher rates of hip fracture, fracture healing problems and bone infections and smoking has been shown to impair soft tissue wound healing. Two new studies, funded by the National Institutes of Health and the National Football League Charities, examined the effects of smoking on fractures and ligament healing in mice and found that healing of both types of injury was delayed. The studies are reported in the December 2006 issue of the Journal of Orthopaedic Research (http://www.interscience.wiley.com/journal/jor), the official journal of the Orthopaedic Research Society.

Led by Hossam B. El-Zawawy of the Washington University School of Medicine in St. Louis, MO the first study involved 35 mice divided into a smoking group, which was exposed to cigarette smoke 6 days per week for a month, and a control group. Surgery was performed on all the mice to achieve a simple fracture. Researchers used type II collagen expression as a marker of cartilage formation (chondrogenesis) during healing. They found that smoking delayed fracture healing and that it began at the early stages of the healing process, although over time it did not inhibit normal healing. Specifically, they were able to show that there was a delay in the development of mature cartilage cells in the mice exposed to cigarette smoke. This was the first study to analyze the molecular and cellular mechanisms of fracture healing in mice exposed to smoke.........

Smoking has been found to contribute to poorer outcomes for people treated for a number of kinds of cancer and now, scientists at Fox Chase Cancer Center have associated smoking and acute side-effects following radiation treatment for prostate cancer. The study was presented today at the 48th Annual Meeting of the American Society for Therapeutic Radiology and Oncology in Philadelphia.

Smoking is linked to an increased risk of radiation-related side effects in cancers of the head and neck, cervix, lung and breast. For this study, scientists analyzed the impact of smoking on gastrointestinal (GI) and genitourinary (GU) side effects for 1,194 prostate cancer patients treated at Fox Chase Cancer Center with 3D conformal radiation treatment between 1991 and 2001. Smoking information collected previous to therapy included status as a current smoker, ex-smoker or non-smoker. Patients treated with androgen deprivation previous to or during therapy were excluded.

"Our patients who smoked during therapy reported having more acute gastrointestinal side-effects such as diarrhea," said Niraj Pahlajani, M.D., lead author on the study and a resident in the radiation oncology department at Fox Chase.

"Fortunately, smoking didn't appear to impact long-term GI side effects or genitourinary side-effects. These results underscore the importance of smoking cessation previous to radiation treatment".........

IMRT versus 3D CRT for prostate cancer
New research findings show men have fewer long-term gastrointestinal side effects with intensity-modulated radiation treatment (IMRT) than with three-dimensional conformal radiation treatment (3D CRT) for prostate cancer therapy, despite the higher doses of radiation used in the IMRT group. These and other data, including long-term genitourinary side effects, were presented today at the 48th Annual Meeting of the American Society for Therapeutic Radiology and Oncology in Philadelphia.

Long-term data comparing 3D CRT for prostate cancer versus the use of IMRT is only now beginning to mature. Few other academic therapy centers have as much experience with 3D CRT and IMRT as Fox Chase.

Both techniques allow precise targeting of the cancer with multiple X-ray beams, but IMRT allows physicians to modulate the radiation dose intensity with far smaller radiation beams. Thus, doses of radiation to the bladder and rectum can be limited. Reducing radiation exposure to healthy normal tissue and other vital organs helps prevent once common side effects such as urinary frequency and diarrhea.

In the study presented today, scientists analyzed data collected prospectively from 1,417 patients treated at Fox Chase. Of these, 928 men were treated with 3D CRT (median follow-up of 63.3 months) and 489 men with IMRT (median follow-up of 29.9 months). The IMRT patients received higher doses of radiation.........
